Miniprep    to extract plasmid

 

Materials:

Methods:

1. Pick glowing colonies of    from experiment 3.1.1, inoculate into of LB Amp. Grow overnight in culture flasks (shake).
2. Follow protocol from Appendix B.1 using of  
3. Perform an analytical gel according to protocol in Appendix E.1.

Results: ( 1/24-1/25/99)

Picked 4 colonies from experiment 3.1.2: 2 from sample , plating volume; 2 from sample , plating volume. Inoculated each into LB Amp flasks. All flasks had growth. Then performed two minipreps using 3 ml each. Resuspended in 100 ml TE.
