Methods:

  1. Follow electroporation protocol in Appendix A.2 using tex2html_wrap_inline3517 tex2html_wrap_inline3068    and tex2html_wrap_inline3521 pGFP(LVA)
  2. Plate on LB Amp plates and grow overnight tex2html_wrap_inline3505 on three different plates using these volumes: tex2html_wrap_inline3525 , tex2html_wrap_inline3467 , and tex2html_wrap_inline3529 .

Results: ( 1/23-1/24/99)

Performed the above protocol twice. The following table shows the number of colonies growing on the plates:

Note: All the colonies did not appear to glow under UV light, so we took 5ml from inoculation performed in next step at put them through FACS. This actually showed high levels of fluorescence, with two peaks. One of the peaks was strongly fluorescent, while the other peak was not. We're assuming that the staining in latter stages will resolve the multiple peaks issue. (also, next time, only take about 1ml for FACS).
