Anneal XbaI-EcoRI Adapter

 

Materials:

Methods:

1. For adapter part ``A'':
   1. Pour 1ml TE in DNA oligo tubes.
   2. Aliquot 100   into sterile tubes: Each tube now contains 86   DNA. freeze all but one .
   3. Dilute one tube 10:1 with TE, to get final volume of 1ml. Concentration: 86  /ml
2. For adapter part ``B'':
   1. Pour 1ml TE in DNA oligo tubes.
   2. Aliquot 100   into sterile tubes: Each tube now contains 96   DNA. freeze all but one .
   3. Dilute one tube 10:1 with TE, to get final volume of 1ml. Concentration: 96  /ml
3. To complete: Aliquot 1.1   (94 ng ``A'', 106 ng ``B'') from each tube into a 0.5 mm thin wall PCR tube.
4. Add TE to final volume of 10  . Put rest of tube .
5. Anneal in PCR machine, with 2 min , then per second, until reach . Hold . Program folder: ``GEO'', Program name: ``ANNEAL''.
