Digest pBR322 and pGFP(LVA) with 2 Enzymes Each

 

Materials:

Methods:

1. thaw pGFP(LVA) and pBR322 in vector form. Also, thaw restriction enzymes, RE Buffers, all on ice.
2. set up the following digests for pBR322 and PGFP(LVA), and make to add the restriction enzymes last:

   

   Here, want to get 0.4   DNA for pBR322 and pGFP(LVA) samples #1, and 0.1   DNA for rest.

3. mix the contents well by gently tapping the bottom of the tube with finger (avoid bubbles). Briefly spin down the contents of the tube in microifuge.
4. Incubate for 1.0 - 1.5 hours
5. Inactivate the restriction enzymes with 15 min
